WE wish to report that reconstituted sperm whale myoglobin prepared by the method of Breslow1 (except that pH 2 was found sufficient to remove all the haem) (I) crystallizes2 in a different habit from those prepared by the method of Rossi-Fanelli et al.3 (II) using haemin of Sigma lot 77B-0220 and our own 57Fe photoporphyrin preparation and the native myoglobin (III). Although all three form type A3 monoclinic prisms, the best developed plane is [001] for II and III, it is [100] for I. There seems to be great interest in reconstituted haemoproteins4,5, so it is important that crystallization habit may be a sensitive test for subtle changes in protein structures.  相似文献

MATURE 5S, 16S and 23S ribosomal RNA species present in E. coli ribosomes are the end products of complex biosyn-thetic pathways. They are formed by reduction in length, and methylation of longer RNA chains transcribed on the ribosomal RNA cistrons of E. coli DNA. Polyacrylamide gel electrophoretic patterns of soluble proteinsfrom intact potato tubers and from wounded tuber tissue wereidentical. An ultrastructural study of life cycle stages of Pneumocystis carinii in infected rat lungs in situ was undertaken utilizing 8 different modes of fixation. Three of the fixatives employed gave good fixation of cysts and intracystic bodies, but for the trophic forms fixation was only fair. Both the trophic forms and intracystic bodies have nuclear pores. The mitochondria of the organism have cristac that appear lamellar. One of the fixation modes revealed a thin, electron-dense layer on the outer surface of the cell wall, a "fuzzy coat" that had not been described previously. This material appears to mediate tight adhesion of trophic forms with other trophic forms, cysts, and with pneumocytcs of the lung alveolus.  相似文献

The activity of the glycolytic enzymes PFK, PFP, PK and aldolaseas well as the content of glucose, fructose, glucose-6-phosphateand fructose-6-phosphate were compared in the embryos of airand oxygen-incubated seeds of Strelitzia juncea. Determinationswere made during the first 4 d of incubation, prior to radicleemergence, which commences on day five for oxygen-treated seeds. No difference in PFK activity was found for the two treatments,and for both treatments PFK tended to increase with the incubationperiod. The fr2, 6P2-stimulated PFP activity was slightly higherfor oxygen-incubated seeds, and showed a significant increasein activity over the 4 d incubation period for both treatments.No significant change in the general trend of PK and aldolaseactivity resulted from incubating the seeds in oxygen. The almost equimolar glucose and fructose contents of the embryoswere lower after 1 d of oxygen incubation of the seeds, andthe content decreased sharply during the incubation period.It is concluded that a moderate increase in the glycolytic capacityof embryos resulted from oxygen treatment of S. juncea seeds. SUMMARY. 1. Pore water chemistry in peaty sediment was monitored for a year at two representative locations of the eutrophic shallow Loosdrecht lakes. The Netherlands. Phosphorus fluxes over the sediment-water interface were calculated using measured concentration gradients in the pore water and compared to fluxes measured under laboratory conditions. Results were analysed with Redundancy Analysis to detect patterns of variation in pore water chemistry and in measured and calculated fluxes, that could be ascribed to environmental variables.
2. It was demonstrated that phosphorus fluxes measured in long-term laboratory incubations were not correlated to any of the pore water characteristics.
3. Initial phosphorus fluxes measured in sediment columns, which varied between −7.7 and 1330 μmol m−2: day−1, were correlated significantly to the calculated phosphorus flux over the sediment-water interface.
4. The high correlation between calculated fluxes of ammonia, phosphorus and methane and measured initial flux of phosphorus, conclusively pointed to mineralization of organic matter as the driving force for phosphorus release from the sediment.
5. Redundancy Analysis demonstrated that the rates of mineralization and phosphorus release were only weakly related to temperature. They appeared to be especially stimulated by the autumnal decrease in temperature which was probably related to an extra input of organic matter.  相似文献

Abstract. Uptake of phytotoxic amounts of metal by higher plants or algae can result in inhibition of several enzymes, and in increase in activity (= induction) of others. Two mechanisms of enzyme inhibition predominate: (1) binding of the metal to sulphydryl groups, involved in the catalytic actionor structural integrity of enzymes, and (2) deficiency of an essential metal in metalloproteins or metal-protein complexes, eventually combined with substitution of the toxic metal for the deficient element. Metal accumulation in the cellular compartment of the enzyme is a prerequisite for enzyme inhibition in vivo. The induction of some enzymes is considered to play a significant role in the stress metabolism, induced by metal phytotoxicity. Peroxidase induction is likely to be related to oxidative reactions at the biomembrane; several enzymes of the intermediary metabolism might be stimulated to compensate for metal-sensitive photosynthetic reactions. The induction of enzymes and metal-specific changes in isoperoxidase pattern can be used as diagnostic criteria to evaluate the phytotoxicity of soils, contaminated by several metals. Lines for future research on metal phytotoxicity are proposed, involving the study of inhibition and induction of enzymes at the different cell membranes (especially the plasmamembrane) in vivo.  相似文献

A single specimen of the early Middle Cambrian (Ordian) orthothecimorph hyolith Gudugu-wan hardmani with preserved gut filling has been discovered in the Montejinni Limestone, Wiso Basin, northern Australia. This represents only the eighth such occurrence worldwide. The specimen is unique among orthothecimorphs in retaining the gut filling in combination with the external mould of the operculum and invites revision of the accepted soft-part recon struction in this group, whereby the opercular lateral processes are now recognized to be ventral. Thus orthothecimorph conchs, if horizontal in life, would have been oriented ventral side down, while hyolithomorphs adopted the reverse orientation: horizontal, ventral side up.  相似文献
